The proposed method is based on the detection and three-dimensional localization of all (resolvable) fluctuations associated with interacting single biomolecules. I leverage interferometric scattering microscopy (iSCAT) to detect the light scattered by a single molecule and achieve microsecond temporal resolution to discern molecule-by-molecule details of protein assemblies and turnover in large biomolecular structures.
