Interactions of intracellular amyloid beta and diagnosis of Alzheimer disease

Alzheimer disease (AD) is one of the most common disorders of elderly. Treatment options for AD especially in later stages are rather limited. Therefore, there is an increasing effort to find new highly specific biomarkers of AD sensitive especially in early stages of the disease. Previously, we evaluated new biomarkers of AD (complexes of amyloid beta peptides and some intracellular proteins) in cerebrospinal fluid. Here, we plan to develop more sensitive methods (ELISA, SPR biosensor) and to detect complexes of oligomeric amyloid or of amyloid beta 1-42 with intracellular binding targets (protein tau, mitochondrial enzymes 17beta-hydroxysteroid dehydrogenase type 10 and cyclophilin D). Values of the new biomarkers will be compared with the current biomarkers (amyloid beta 1-42, tau and phospho-tau) and with the degree of cognitive impairment. New biomarkers are expected to improve an ability to discriminate either AD from other types of dementia either mild cognitive impairment due to AD from that caused by other ethiology.
