Circulating exosomes have emerged as promising biomarkers for a variety of diseases. Exosomes offer two types of features that may be targeted: membrane proteins expressed at the exosome surface and cargo molecules (most often proteins and miRNAs) encapsulated inside the exosomes. The current methodologies mostly target either intact exosomes for direct exosome detection/profiling via membrane proteins or lysed exosomes for molecular analysis of their total protein, miRNA/mRNA or lipid content. In the proposed project, we shall develop a new biosensing methodology that will enable simultaneous detection and quantification of exosomes and exosomal cargo molecules to take full advantage of the richness of information embraced in exosomes.
